Simon Kirke and Lorraine Kirke welcomed Domino into the world on December 17, 1983, in London, England. Her mother is a fashion designer and the proprietor of a store, while her father plays drums and is a part of the renowned rock bands Bad Company and Free. Together with her sisters, actors Lola Kirke and Jemima Kirke, Domino grew up in New York City.

Domino first began singing and playing the piano when she was a young child. Later, she pursued classical vocal studies at New York City's Fiorello H. LaGuardia High School of Music, Art, and Performing Arts. She obtained a performing arts degree while attending Bard College as well.

In a New York City pub in 2014, Domino and Penn Badgley first connected. They soon began dating, and in 2016 they became engaged. The pair wed in a courthouse ceremony in Brooklyn, New York, in February 2017.
